蛋白为什么要冻干？冻干对蛋白的影响有哪些？

蛋白质对热敏感，冻干能使绝大部分蛋白质的活性保留下来，提高蛋白的稳定性并延长保存时间，同时降低运费。冻干有可能会造成蛋白的活性部分损失，聚集和其它变性问题。但可以通过添加保护剂（稳定剂，添加剂，辅料）和控制冻干的各种条件来尽可能降低这些负面的影响。

冻干前为什么向蛋白溶液中加保护剂？一般冻干保护剂有哪几种？你们的产品通常加的保护剂是什么？

保护剂是用来在冻干和储存过程中保护蛋白的。常用的保护剂或稳定剂有糖类，多元醇，聚合物，表面活性剂，某些蛋白和氨基酸等。我们通常加8%（质量比体积）的海藻糖和甘露醇作为冻干保护剂。
海藻糖可明显阻止蛋白质二级结构改变以及冻干过程中蛋白质的伸展和聚集。
甘露醇也是一种普遍应用的冻干保护剂和填充剂。有文献报道甘露醇可以降低某些蛋白的冻干后聚集情况。

怎样复溶蛋白干粉？我应该加无菌水还是PBS来复溶？

我们建议使用过滤后的无菌纯化水作为复溶剂。添加纯化水至推荐的体积，轻轻摇晃西林瓶，使蛋白溶解完全均匀。切忌剧烈摇晃。PBS或者其它缓冲液也可以用来做复溶剂，因为PBS中的盐可以在体积微量的情况下忽略不计。如果可能，我们建议做一下两种复溶剂的效果比较。
